Establishment & Study of Mutant Mouse with Brachyclinodactyly in the ENU Mutagenesis Screening

Abstract

|
ENU(N-ethyl-N-nitrosourea) is one of the most effective chemical mutagens. 200 mg/kg ENU was injected into peritoneum of male BALB/c for spermatogonial mutagenization. Mutant mouse showing digit dysmorphology was initially screened in the 3rd generation after ENU injection. The mutant could be grossly distiguished from normal littermates at birth. The dysmorphology of the 1st, 2nd and 5th toes in all feet was characteristic. They were shortened or crooked. In carrrier & carrier mating, the incidence rate was 28.7% in the observed 228 mice until G9 generation. In carrier and mutant mating, it was 47% in the observed 86 mice. For the confirmation of genetic mode, breeding experiment was performed between mutant carrier and normal C57BL/6. No mutants were observed in F1 generation but 39 mutants(male/female = 20/19) out of 171 progenies were produced in F2. The sex ratio of mutants was not significantly different. Therefore it was supposed that the responsible gene was located on autosomal chromosome and transmitted next generation by recessive mode. This mutant phenotype, brachydactyly and clinodactyly was considered to be single gene effect in the base of mendelian law.
